目的:研究针刺疗法联合塞来昔布对老年类风湿性关节炎患者的临床疗效及其对患者血清基质金属蛋白酶-3(MMP-3)、白细胞介素-6(IL-6)及自身抗体水平的影响。方法:选取2014年10月至2015年9月本院收治的86例类风湿性关节炎患者,随机分为观察组(43例)和对照组(43例)。对照组使用塞来昔布胶囊治疗,观察组在对照组基础上联合针刺疗法。分析和比较两组患者临床疗效、治疗前后20 m步行时间、晨僵时间、关节压痛数、治疗前后MMP-3、IL-6及自身抗体水平的变化。结果:观察组总有效率显著高于对照组[90.70%(39/43)比65.12%(28/43)](P0.05)。治疗后,两组患者20m步行时间、晨僵时间、关节压痛数均较治疗前显著降低或缩短,其中观察组明显低于或短于对照组(P0.05)。治疗后,两组患者血清MMP-3、IL-6水平较治疗前显著降低,其中观察组显著低于对照组(P0.05)。治疗后,两组患者AFA、抗-CCP抗体、抗Sa抗体、AKA、ANA自身抗体阳性率较治疗前显著降低,其中观察组显著低于对照组(P0.05)。结论:针刺疗法联合塞来昔布治疗老年类风湿性关节炎的临床疗效良好,能有效降低患者血清MMP-3、IL-6水平及自身抗体阳性率。  相似文献

目的:探讨百令胶囊对多囊卵巢综合征患者卵泡液骨形态蛋白(BMP)、生长分化因子(GDF)-9及胰岛素样生长因子(IGF)的影响。方法:选择2013年7月至2016年7月我院接诊的90例多囊卵巢综合征患者,通过随机数表法分为观察组(n=45)和对照组(n=45),对照组使用达英-35治疗,观察组在此基础上使用百令胶囊。比较两组糖代谢指标、内分泌指标、卵泡液BMP-15、GDF-9、IGF-1水平的变化以及临床疗效。结果:治疗后,观察组空腹血糖(FBG)、空腹胰岛素(FINS)、胰岛素抵抗(HOMA-IR)、血清卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)水平水平均显著低于对照组(P0.05),卵泡液BMP-15、GDF-9水平显著高于对照组,IGF-1水平明显低于对照组(P0.05),总有效率显著高于对照组(P0.05)。结论:百令胶囊质量多囊卵巢综合征效果显著,可有效调节内分泌,并改善卵泡液BMP-15、GDF-9、IGF-1的水平。  相似文献

目的:探讨二甲双胍和塞来昔布单独或联合应用对胰腺癌细胞增殖和Caspase-3活性的影响。方法:体外培养人胰腺癌BxPC-3和As PC-1细胞,用四甲基偶氮唑盐(MTT)法检测不同浓度二甲双胍和塞来细胞对胰腺癌细胞存活率的影响。联合实验分4组:对照组、二甲双胍组(MET,15 mmol/L)、塞来昔布组(CEL,100μmmol/L),二甲双胍和塞来昔布联合组(MET 15 mmol/L+CEL100μmmol/L),孵育48 h,用MTT检测细胞存活率,用Caspase-3比色测定试剂盒测定Caspase-3活性。结果:二甲双胍和塞来昔布单药均可以时间和剂量依赖性方式降低胰腺癌Bx PC-3和As PC-1细胞的生存率。两种细胞系的各加药组细胞存活率与对照组比较差异均存在统计学意义(P0.01),联合实验组的细胞存活率明显低于单独用药组(P0.01)。二甲双胍和塞来昔布单药处理的胰腺癌Bx PC-3和As PC-1细胞Caspase-3活性均显著高于对照组(P0.01),二甲双胍和塞来昔布联合实验组Caspase-3活性均明显高于单药处理组和对照组(P0.01),但二甲双胍组和塞来昔布组之间的Caspase-3活性比较差异无统计学意义(P0.05)。结论:二甲双胍和塞来昔布联合作用可协同抑制胰腺癌细胞的增殖,并通过激活Caspase-3活性促进胰腺癌细胞的凋亡,其联合应用可能成为胰腺癌药物治疗的有效策略。  相似文献

目的:采用荧光定量PCR法检测前列腺癌循环miRNA-141表达,并研究五苓胶囊对其影响。方法:选取我院泌尿外科收治的前列腺癌患者20例,随机分为A、B两组,各10例;A组给予常规治疗,B组在常规治疗基础上给予加用五苓胶囊治疗。选取同期住院前列腺增生患者20例,正常对照组为同时期体检健康者20例。采用荧光定量PCR法检测正常对照组、前列腺增生组以及前列腺癌组血清miRNA-141的表达情况,以及前列腺癌组治疗前TPSA(前列腺特异性抗原)含量。结果:前列腺癌组的血清miRNA-141的表达水平为正常对照组的(11.22±8.19)倍,明显高于前列腺增生组(1.65±1.73)和正常对照组,差异有统计学意义(P0.05);前列腺增生组和正常对照组比较无明显差异(P0.05);前列腺癌组血清miRNA-141的表达水平与TPSA水平呈正相关关系,相关系数r=0.753,有统计学意义(P0.05)。治疗后,前列腺癌A组与B组的血清miRNA-141的表达水平均有所改善,与A组(5.82±4.51)相比较,B组的miRNA-141的表达水平(1.93±0.67)明显降低,差异有统计学意义(P0.05)。结论:前列腺癌患者血清中miRNA-141表达水平明显升高,且与TPSA水平呈正相关关系,五苓胶囊能够降低miRNA-141表达。  相似文献

Non-steroidal anti-inflammatory drugs (NSAIDs) achieve their anti-inflammatory effect by inhibiting cyclooxygenase activity. We previously suggested that in addition to cyclooxygenase-inhibition at the gastric mucosa, NSAID-induced gastric mucosal cell death is required for the formation of NSAID-induced gastric lesions in vivo. We showed that celecoxib exhibited the most potent membrane permeabilizing activity among the NSAIDs tested. In contrast, we have found that the NSAID rofecoxib has very weak membrane permeabilizing activity. To understand the membrane permeabilizing activity of coxibs in terms of their structure–activity relationship, we separated the structures of celecoxib and rofecoxib into three parts, synthesized hybrid compounds by substitution of each of the parts, and examined the membrane permeabilizing activities of these hybrids. The results suggest that the sulfonamidophenyl subgroup of celecoxib or the methanesulfonylphenyl subgroup of rofecoxib is important for their potent or weak membrane permeabilizing activity, respectively. These findings provide important information for design and synthesis of new coxibs with lower membrane permeabilizing activity.  相似文献

Alginate capsules containing anionic complex silverthiosulfate (STS) [Ag(S2O3)2 3-] were placed in the culture tubes over minimal growth media for studying whether STS could be used at higher concentrations to sustain ethylene-inhibiting effect during conservation of microplants in six potato (Solanum tuberosum L.) genotypes in vitro. Different concentrations of STS (0.1, 0.5, 1.0, 2.0, 3.0 and 4.0 mM) were incorporated into the alginate capsules, and 12 alginate-STS capsules were tested in semisolid (7 g l–1 agar) minimal growth medium containing 20 g l–1 mannitol and 40 g l–1 sucrose. This indirect supplementation of STS through alginate capsules rendered reduced total availability of STS in the minimal growth medium as compared to when it was directly supplemented in the medium at a given concentration. Growing of microplants in the presence of alginate-STS capsules improved the microplant growth and reduced the culture abnormalities over a period of 16 months under minimal growth conditions. Most significant improvement in microplant growth was in terms of green leaf production and leaf senescence. Vitrification, flaccidity and other growth abnormalities, viz., leaf loss, abnormal stem swelling and necrosis were not observed when the microplants were conserved in the presence of alginate-STS capsules. To foster optimum microplant growth and reduce culture abnormalities, potato microplants could favourably be maintained in the presence of 0.5–1.0 mM alginate-STS capsules during minimal growth conservation. Higher concentrations of alginate-STS capsules (>1.0 mM) were in general detrimental to potato microplant growth and survival during prolonged storage in vitro. Release kinetics of STS from the alginate-STS capsules, its distribution in the medium and accumulation of silver in potato microplants were studied using 110mAg. The release rate of STS from the capsules was found to be directly proportional to the concentrations of alginate-STS capsules. A distinct concentration gradient of 110mAg in the medium with increasing depth from top to bottom, and its accumulation in the potato microplants may be attributed to the improved anti-ethylene action of STS at higher concentrations through alginate capsules.  相似文献

Dictyostelium discoideum is a promising eukaryotic host for the expression of heterologous proteins requiring post‐translational modifications. However, the dilute nature of D. discoideum cell culture limits applications for high value proteins production. D. discoideum cells, entrapped in sodium cellulose sulfate/poly‐dimethyl‐diallyl‐ammonium chloride (NaCS‐PDMDAAC) capsules were used for biosynthesis of the heterologous protein, soluble human Fas ligand (hFasL). Semi‐continuous cultivations with capsules recycling were carried out in shake flasks. Also, a scaled‐up cultivation of immobilized D. discoideum for hFasL production in a customized vitreous airlift bioreactor was conducted. The results show that NaCS‐PDMDAAC capsules have desirable biophysical properties including biocompatibility with the D. discoideum cells and good mechanical stability throughout the duration of cultivation. A maximum cell density of 2.02 × 107 cells mL?1 (equivalent to a maximum cell density of 2.22 × 108 cells mL?1 in capsules) and a hFasL concentration of 130.40 μg L?1 (equivalent to a hFasL concentration of 1434.40 μg L?1 in capsules) were obtained in shake flask cultivation with capsules recycling. Also, a maximum cell density of 1.72 × 107cells mL?1 (equivalent to a maximum cell density of 1.89 × 108 cells mL?1 in capsules) and a hFasL concentration of 106.10 μg L?1 (equivalent to a hFasL concentration of 1167.10 μg L?1 in capsules) were obtained after ~170 h cultivation in the airlift bioreactor (with a working volume of 200 mL in a 315 mL bioreactor). As the article presents a premier work in the application of NaCS‐PDMDAAC immobilized D. discoideum cells for the production of hFasL, more work is required to further optimize the system to generate higher cell densities and hFasL titers for large‐scale applications. © 2014 American Institute of Chemical Engineers Biotechnol. 目的:探究银灵通胶囊对脂质代谢的影响及其机制。方法:建立大鼠高脂模型,用药后检测其血脂、丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活性,检测肝脏组织的高密度脂蛋白受体SR-BI、低密度脂蛋白受体(LDLR)、氧化低密度脂蛋白(ox-LDH)受体CD36蛋白表达的mRNA表达水平,检测血管组织学变化。结果:高脂饮食明显升高大鼠血清中总胆固醇(CH)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和动脉硬化指数(AI值),银灵通胶囊组可降低上述指标,且呈一定的浓度依赖;高脂饮食可增加肝脏中SR-BI及CD36表达,降低LDLR表达,银灵通胶囊引起SR-BI的过度表达,使LDLR表达增加,CD36表达下降。高脂饮食使血清中MDA的含量增加,给予银灵通胶囊后,明显降低血清MDA的含量。结论:银灵通胶囊具有调节脂质代谢,抗动脉粥样硬化(AS)及抗脂质过氧化作用。其机制与银灵通胶囊能引起肝脏中SR-BI的过度表达及LDLR表达增加,降低肝脏中CD36表达和血清MDA含量有关。  相似文献

目的 探讨双歧三联活菌胶囊联合参苓白术散对腹泻型肠易激综合征(IBS)患者血浆生长抑素(SS)和神经肽Y(NPY)水平影响及疗效观察.方法 选择64例腹泻型IBS患者,随机分为观察组与对照组.两组患者均予以双歧三联活菌胶囊3粒/次,2次/d.观察组在此治疗基础上加用中药方参苓白术散,1剂/d,两组疗程均为6周.观察并比较治疗前后血浆SS和NPY水平的变化,并分析治疗后临床效果及不良反应.结果 治疗6周后,两组患者血浆SS水平较治疗前明显下降(P<0.05和P<0.01),NPY水平较治疗前明显上升(P<0.05和P<0.01),观察组下降或上升幅度明显大于对照组(P<0.05);观察组治疗总有效率高于对照组(x2=4.27,P<0.05),观察组患者治疗期间发生3例药物不良反应,对照组发生1例,症状均较轻,两组比较差异无统计学意义(x2=0.27,P>0.05).结论 双歧三联活菌胶囊联合参苓白术散治疗腹泻型IBS效果良好,安全性较好,其机制可能与其降低血浆SS水平,升高血浆NPY水平密切相关.  相似文献
